复合酶水解蚯蚓蛋白的研究

    摘要:

    蚯蚓是一种富含蛋白质的功能性生物资源,为提高蚯蚓蛋白水解度,对水解蚯蚓复合酶配比进行优化,并建立复合酶配比优化回归方程模型;采用U30(3013)均匀设计试验对复合酶(木瓜蛋白酶、酸性蛋白酶、菠萝蛋白酶)配比进行设计,以蚯蚓蛋白水解度为指标,筛选出水解度最高的复合酶配比;结果表明,配比优化回归方程为:Y=58975+8270x2+12256x1x2x3+7677x1x3-5243x22,回归方程高度显著(P <005);分析预测最佳配比为:木瓜蛋白酶232%、酸性蛋白酶537%、菠萝蛋白酶231%,模型预测最大水解度为6260%,实际验证水解度为(6284±021) %,高于试验组与模型预测值说明配比优化后的复合酶可提高酶解效率并为蚯蚓蛋白酶解工艺提供参考。

    Abstract:

    Earthworm is a kind of functional biological resource rich in protein.In order to increase the degree of protein hydrolysis of earthworm, the ratio of compound enzyme was optimized, and the regression equation model of compound enzyme ratio was established.U30 (3013) uniform design experiment was used to design the ratio of complex enzymes (papain, acidic proteinase, bromelain), and the ratio of the highest degree of hydrolysis of earthworm protein was selected as the index.The results showed that the ratio optimization regression equation was Y=58975+8270x2+12256x1x2x3+7677x1x3-5243x22, and the height of the regression equation was significant (P < 005).The optimal ratio was as follows:232% papain, 537% acidic protease and 231% bromelain.The value of model predicted the maximum hydrolysis degree to be 6260%, and the actual test showed that the hydrolysis degree was (6284±021) %, higher than the experimental group and the model predicted.Therefore, the optimized ratio of complex enzymes can improve the efficiency of enzymatic hydrolysis and provide a reference for earthworm proteolytic process.
