    摘要:

    通过平板划线法从猪粪废水中分离纯化得到一株产氢光合细菌PSB〖KG-*2〗-〖KG-*6〗ZF1,对其菌体进行染色观察、生理生化试验以及活细胞吸收光谱的测定,初步鉴定菌属于红螺菌科红螺菌属(〖WTBX〗Rhodospirillum〖WTBZ〗)光合细菌;对菌培养过程中的营养条件实验分析后获得的最优培养基配方为:乙酸钠2.0 g,氯化铵1.0 g,碳酸氢钠0.5 g,氯化钠1.0 g,磷酸二氢钾0.2 g,六水氯化镁0.2 g,T.M储液1 mL,酵母膏0.8 g,蒸馏水1 000 mL;考察环境因素对菌种生长的影响,发现光合细菌PSB〖KG-*2〗-〖KG-*6〗ZF1在搅拌状态下,接种量为10%,光照强度为3 000 lux,溶解氧量为2.02 mg/L,初始pH为7.0时生长状态最佳;获得高浓度的PSB〖KG-*2〗-〖KG-*6〗ZF1菌液有利于后续产氢实验。

    Abstract:

    The photosynthetic bacteria PSBZF1,identified as Rhodospirillum by bacteria dyeing,physiological and biochemical test and absorption spectrum test of live cells,was acquired in porcine dung by means of flat lining separation. Through experiment analysis of its nutrition condition in the process of cultivation,the best composition of culture medium was received as follows: sodium acetate 2.0g,ammonium chloride 1.0g,sodium bicarbonate 0.5g,sodium chloride 1.0g,potassium dihydrogen phosphate 0.2g,magnesium chloride hexahydrate 0.2g,T.M storage solution 1.0mL,yeast cream 0.8g,and distilled water 1000mL. By examining the influence of environment factors on the bacteria growth,the best growth condition for PSBZF1 was obtained as follows: inoculums of 10%,illumination intension of 3000lux,dissolved oxygen 2.02mg/L,initial pH 7,and under stirring condition. High concentration of PSBZF1 culture is conducive to continuous hydrogen production experiment.
